Navigating the Post–Triple-Class Era: Promising New Drugs and Treatment Approaches

Patients with relapsed or refractory multiple myeloma who have been exposed to proteasome inhibitors, immunomodulatory agents, and anti-CD38 monoclonal antibodies represent a growing population with complex therapeutic needs. This session will review the rapidly evolving treatment landscape in the post–triple-class era, highlighting emerging therapies that are redefining outcomes for heavily pretreated patients. Faculty will discuss recent clinical advances in BCMA- and GPRC5D-targeted immunotherapies, including bispecific antibodies, antibody-drug conjugates, and cellular therapies, as well as novel cereblon E3 ligase modulators (CELMoDs). Key data from pivotal global clinical trials and major hematology meetings will be examined, with a focus on treatment sequencing, patient selection, toxicity management, and strategies to optimize durable responses in contemporary myeloma.
