Bispecifics and Trispecifics in Action: Translating Clinical Advances into Everyday Myeloma Practice

Bispecific antibodies have rapidly transformed the treatment landscape of relapsed and refractory multiple myeloma, while next-generation trispecific constructs are poised to further expand immune-based therapeutic options. This session will review the latest clinical advances in BCMA- and GPRC5D-directed bispecific therapies, including pivotal randomized trials that are moving these agents into earlier lines of treatment and demonstrating activity in high-risk populations such as extramedullary disease. Faculty will discuss emerging trispecific approaches designed to address antigen escape and enhance T-cell engagement. Practical considerations for everyday clinical practice—including patient selection, sequencing strategies, infection prevention, toxicity management, treatment logistics, and quality-of-life outcomes—will be emphasized. Attendees will gain evidence-based insights into integrating these novel immune therapies into contemporary myeloma care.
